I. Overcoming Challenges in Developing a Scalable Process for Protein Therapeutics.
There are many challenges in developing a successful scalable process for protein therapeutics. Some of these are: efficiency, reproducibility, cost-effectiveness, high quality, and speed to clinic. Suggestions, for developing successful strategies to overcome all these, will be discussed. There will be an emphasis on systematic methods to successfully refold proteins and optimizing chromatography.

II. The Use of Dual Salts to Improve Capacity and Selectivity in HIC and Mixed-Mode Resins as a Logical Extension to HIC Applications.
Hydrophobic interaction chromatography (HIC) is a unique mode that offers several advantages over ion exchange and size exclusion chromatography. HIC is often used to remove aggregates, host cell proteins and leached Protein A. Recent advances have shown that using a dual salt combination (mixing kosmotropic and chaotropic salts) can enhance the binding capacity of the resin, aid in protein solubility and in some cases improve the selectivity of the HIC resin being used. These factors are critical to success in process chromatography applications and this strategy of combining different salts in HIC will be discussed. In Part II of this talk, Mixed-Mode Chromatography (MMC) often employs HIC as one of the modes utilized which enhances the selectivity. MMC, therefore, can be thought of as a logical extension to HIC. MMC often has even higher capacities with unique selectivities when compared to HIC resins. The newly introduced TOYOPEARL MX-Trp-650M resin has very high capacity and excellent salt tolerance under common mixed-mode chromatographic conditions. These unique features along with some of the other properties of TOYOPEARL MX-Trp-650M will be discussed.
